Using iRT, a Normalized Retention Time for More Targeted Measurement of Peptides

Proteomics Scheduling targeted proteomics acquisition based upon precise peptides’ retention time (RT) improves quantitative accuracy and multiplexing. Here the authors presented the indexed RT (iRT) concept, a dimensionless value for RT prediction. The iRT value is based upon a set of so-called iRT-peptides which are used as internal RT standards. Using iRT improves quantification by narrowing the scheduled acquisition windows and facilitates RT normalization across laboratories and chromatography methods. Later, that concept was transferred by the authors to the targeted extraction from DIA/SWATH data.
